Description of Target:
Beta-catenin is an adherens junction protein. Adherens junctions (AJs; also called the zonula adherens) are critical for the establishment and maintenance of epithelial layers, such as those lining organ surfaces. AJs mediate adhesion between cells, communicate a signal that neighboring cells are present, and anchor the actin cytoskeleton. In serving these roles, AJs regulate normal cell growth and behavior. At several stages of embryogenesis, wound healing, and tumor cell metastasis, cells form and leave epithelia. This process, which involves the disruption and reestablishment of epithelial cell-cell contacts, may be regulated by the disassembly and assembly of AJs. AJs may also function in the transmission of the 'contact inhibition' signal, which instructs cells to stop dividing once an epithelial sheet is complete.Beta-catenin is an adherens junction protein. How do Antibody Verify’s reagents play a role in your experimental goals?


Our lab is interested in blocking pathways that are mis-regulated in cancer and inflammatory diseases by using small molecule inhibitors. One such pathway is the wnt signaling pathway, which we have developed a drug for. By using antibodies to proteins that are key players, we want to elucidate the mechanisms that are affected by the inhibition of this pathway.


How many different experimental trials were conducted using the antibody sample?


I have tried the ChIP assay and western blots twice


What type of experimental sample are you using and how did you prepare it?


For my experiments I used a human colon cancer cell line, HCT116. These are loosely adherent cells grown in 15-cm tissue culture dishes. For ChIP, cells were prepared using the Farnham protocol. For westerns, cells were lysed using the M-Per cell extraction kit from Pierce.


What applications did you test the antibody in? Please include dilutions of the primary and secondary reagents.


For ChIP, I use 2 micrograms of antibody. For westerns, the antibodies were diluted 1:1000 as specified by your product sheet. Secondary anti-rabbit IgG-HRP was used at a dilution of 1:10,000.



What controls were used in your experiment? Please include your positive control.



For ChIP, antibodies to CBP and p300 were used as positive controls. Normal igG was used as the negative control. For western a commercial beta-catenin antibody from BD was used as the positive control


How did you store the antibody after re-suspension?


The antibodies were stored at -20 o C


Please provide the protocol for your application procedure. Please be as detailed as possible.


For ChIP, please see the Farnham protocol attached. For western, SDS-PAGE was performed and proteins were transferred onto PVDF overnight at 4 o C at 30 volts. After transfer, the membrane is blocked with 5% nonfat milk in TBST. Primary antibody was added at the appropriate dilutions (see attached results) and incubated overnight at 4 o C. After thorough washing, the membrane is incubated in secondary antibody conjugated to HRP for 1 hour at room temperature. The membrane is then exposed to film using ECL substrate after 30 minutes of washing.




How would you rate this antibody on a scale from 1-5? Why?



For ChIP, the AAS01006C_P050 performed very well compared to a commercial beta-catenin antibody. AAS00006E and AAS01140E, however, did not perform well in ChIP. All 3 antibodies performed well in western blots compared to a well- known commercial antibody. I would rate AAS01006C_P50 a 4 on ChIP and 3 on western. I would rate AAS00006E and AAS01140E a 1 on ChIP and 4 on western



Would you use this antibody in future experiments?



I would definitely use AAS01006C_P050 for ChIP and the other two for western blotting




Do you believe the information about the reagent on Antibody Verify’s website is correct?



Yes, I believe the info is correct since the antibodies worked well in my experiments.



If the antibody works, do you plan to use it in future experiments or to publish your data? Why or why not?



Yes, I will definitely use the AAS01006C_P050 antibody for future ChIP assays since it outperformed an antibody that is commercially available. I would use the other two antibodies for westerns since it did equally well if pricing is competitive.
